چکیده

In macroscopic organisms, aging is often obvious; in single-celled organisms, where there is the greatest potential to identify the molecular mechanisms involved, identifying and quantifying aging is harder. The primary results in this area have come from organisms that share the traits of a visibly asymmetric division, and an identifiable juvenile phase. As reproductive aging must require a differential distribution of aged and young components between parent and offspring, it has been postulated that organisms without these traits do not age, thus exhibiting functional immortality. Through automated timelapse microscopy, we followed repeated cycles of reproduction by individual cells of the model organism Escherichia coli, which reproduces without a juvenile phase and with an apparently symmetric division. We show that the cell that inherits the old pole exhibits a diminished growth rate, decreased offspring production, and an increased incidence of death. We conclude that the two supposedly identical cells produced during cell division are functionally asymmetric; the old pole cell should be considered an aging parent repeatedly producing rejuvenated offspring. These results suggest that no life strategy is immune to the effects of aging, and therefore immortality may be either too costly or mechanistically impossible in natural organisms.

An aging-independent replicative lifespan in a symmetrically dividing eukaryote

The replicative lifespan (RLS) of a cell-defined as the number of cell divisions before death-has informed our understanding of the mechanisms of cellular aging. However, little is known about aging and longevity in symmetrically dividing eukaryotic cells because most prior studies have used budding yeast for RLS studies. Here, we describe a multiplexed fission yeast lifespan micro-dissector (m...

Aging may be a conditional strategic choice and not an inevitable outcome for bacteria.

Aging is known in all organisms that have different somatic and reproductive cells or in unicellular organisms that divide asymmetrically. Bacteria that divide symmetrically were believed to be immune to natural aging. The demonstration of functionally asymmetric division and aging in Escherichia coli recently has challenged this belief and led to the suggestion that aging might be inevitable f...

Robust Growth of Escherichia coli

The quantitative study of the cell growth has led to many fundamental insights in our understanding of a wide range of subjects, from the cell cycle to senescence. Of particular importance is the growth rate, whose constancy represents a physiological steady state of an organism. Recent studies, however, suggest that the rate of elongation during exponential growth of bacterial cells decreases ...

Using Taguchi Experimental Design to Develop an Optimized Synthesis Procedure of Sodalite Prepared by Microwave and Ultrasonic Assisted Aging

Perlite or pearl stone is a natural glass which has formed by rapid cooling of viscous lava or magma. Perlite is mainly composed of silica, aluminum, potassium and sodium. In this experiment, perlite was used as a low-cost source of Si and Al to synthesis of sodalite zeolite using the alkaline hydrothermal treatment. A statistical Taguchi design of experiments was employed to evaluate the effec...
